Most Harley-Davidson touring motorcycles have a separate sump for the engine, transmission, and primary chaincase (hence the 3-holes). AMSOIL recommends 20W-50 Synthetic V-Twin Motorcycle Oil for all three in most Harleys.
In order to do a 3-hole oil change, you'll have to purchase additional O-rings and more 20W-50 v-twin oil since the V-Twin oil change kits only include enough oil for just the engine.

20W-50 oil is a commonly used oil weight for many different applications, including V-Twin engines (like Harleys - where it can be used in all 3 sumps), it's great for high-mileage diesel and gasoline engines, also recommended in classic car engines with flat-tappet camshafts, excels in hydrostatic transmissions, and is also used in high-performance competition diesel engines.
For maximum protection, you should use AMSOIL V-Twin Primary Fluid which will NOT void your warranty. You can also use AMSOIL V-Twin Motorcycle Oil which was designed to be used as an engine oil, transmission, and primary fluid. Check here to find the best oil for your specific Harley.
If also using an AMSOIL motorcycle oil filter, you can run your motorcycle up to twice the manufacturer-recommended drain interval with AMSOIL in the engine and transmission (and primary chaincase for V-Twins) or up to one year, whichever comes first.
The best all-around motor oil for Harley's is AMSOIL V-Twin Motorcycle Oil which can also be used as primary fluid and transmission fluid.

You can mix synthetic and regular oil in a motorcycle, but it is not recommended. Mixing synthetic and conventional oil degrades the lubricating properties and chemical additives in synthetics.

The term metric motorcycle refers to motorcycles built outside of the US, brands like Honda, Yamaha and Suzuki. Most of these non-US brands measure their engines in cubic centimeters (CCs) rather than cubic inches.
